Overview

CJC-1295 with DAC incorporates a drug affinity complex — a maleimidoproprionic acid (MPA) moiety that forms a covalent bond with cysteine-34 on circulating albumin. This albumin conjugation dramatically extends the half-life to 6–8 days, enabling once-weekly or twice-weekly research dosing protocols. The trade-off is a blunted, non-pulsatile GH elevation rather than the sharp physiological pulses produced by the no-DAC variant.

Mechanism of Action

After subcutaneous injection, the reactive MPA group on CJC-1295 DAC covalently binds to albumin's free thiol group (Cys-34). The albumin-bound peptide circulates for days, continuously stimulating GHRH receptors and producing sustained GH elevation. This results in chronically elevated IGF-1 levels. The sustained (non-pulsatile) GH pattern differs fundamentally from physiological GH secretion and from the no-DAC variant, which has implications for receptor desensitization and downstream signaling in long-term research models.

Technique

Inject solvent slowly down the vial wall. Swirl gently until dissolved. Avoid vigorous agitation.

Notes

The DAC moiety is reactive — avoid contact with thiol-containing compounds or reducing agents in solution, as these may compete for the maleimide group prior to albumin binding.
