Overview

Selank (Thr-Lys-Pro-Arg-Pro-Gly-Pro) is a synthetic heptapeptide analogue of the endogenous immunomodulatory tetrapeptide tuftsin (Thr-Lys-Pro-Arg). The Pro-Gly-Pro extension, identical to that in Semax, confers metabolic stability. Developed at the Institute of Molecular Genetics in Russia, Selank has been approved in Russia as an anxiolytic and nootropic agent. It is studied for its GABAergic modulation, enkephalinase inhibition, and cytokine-regulatory properties.

Mechanism of Action

Selank modulates GABAergic neurotransmission, producing anxiolytic effects without the sedation or dependence associated with benzodiazepines. It inhibits enkephalin-degrading enzymes (enkephalinase), prolonging endogenous opioid peptide activity and contributing to anxiolytic and mood-stabilizing effects. Selank also regulates IL-6 expression and T-helper cell balance, linking its anxiolytic and immunomodulatory properties. In memory models, it enhances consolidation and retrieval, possibly through BDNF upregulation and cholinergic pathway modulation.

Notes

Like Semax, Selank is also studied via intranasal administration. For intranasal research protocols, sterile saline is the preferred vehicle.

Russian clinical use: 400 mcg intranasal (200 mcg per nostril), 2–3 times daily.
